ASCENT: Sacituzumab govitecan versus physician's-choice chemotherapy in previously treated metastatic triple-negative breast cancer

Breast Cancer · Medical · Relapsed / Refractory · 529 randomized; primary efficacy population 468 patients

Treatment: Sacituzumab govitecan 10 mg/kg IV on days 1 and 8 every 21 days versus physician's-choice single-agent chemotherapy.

What did it show? Final analysis: median PFS 4.8 vs 1.7 months (HR 0.41, 95% CI 0.33–0.52); median OS 11.8 vs 6.9 months (HR 0.51, 95% CI 0.42–0.63). These median differences are not NNTs.

Critical clinical review
Sacituzumab govitecan substantially improved PFS and OS in previously treated metastatic TNBC. Grade ≥3 neutropenia and diarrhoea are clinically important; monitor for febrile neutropenia and manage diarrhoea proactively. The primary efficacy population excluded active brain metastases.

How to read the evidence

Every entry should make the clinical consequence easier to see.

01 · What the trial reported

HR, RR, OR or another relative measure, exactly as reported.

02 · What changes for patients

Absolute difference at a defined time point or clinically meaningful unit.

03 · Put it in 100 patients

NNT/NNH when appropriate, always tied to a specified endpoint and time horizon.
