Study design, population and eligibility

Design and sample

Phase II open-label single-arm multicohort basket study.

Population

Advanced solid tumours with FGFR1–FGFR3 fusions/rearrangements or activating mutations.

Treatment and comparator

Intervention: Pemigatinib 13.5 mg daily, 2 weeks on/1 week off.

Comparator: No randomized comparator; molecular cohorts.

Endpoints

ORR in cohorts A and B primary.

Key efficacy and clinical outcomes

ORR 26.5% in fusion/rearrangement cohort (13/49), 9.4% mutation cohort (3/32), 3.8% kinase-domain/uncertain cohort (1/26); median PFS 4.5 and 3.7 mo in A/B.

Median survival and hazard ratios are not fixed-time event risks; NNT is not inferred from them.

Toxicity and treatment burden

Hyperphosphataemia 84%, stomatitis 53%; ocular/nail/skin toxicity.

Absolute effect and NNT/NNH

Single-arm basket; response is alteration- and tumour-context-dependent; no comparative effect.

NNT/NNH is not calculated unless a directly reported, comparable fixed-time risk difference supports it. Median survival and hazard ratios alone do not provide an NNT.

Critical interpretation and limitations

Single-arm basket; response is alteration- and tumour-context-dependent; no comparative effect.
