CheckMate 816

Does nivolumab plus neoadjuvant chemotherapy improve survival in resectable NSCLC?

Population and treatment

358 patients with resectable stage IB–IIIA NSCLC.

Intervention vs comparator: Three cycles nivolumab + platinum chemotherapy vs chemotherapy alone, followed by surgery.

Results

At median follow-up 68.4 months, 5-year OS 65.4% vs 55.0%; HR 0.72 (95% CI 0.523–0.998; p=0.048).

Toxicity and treatment burden

55

Full grade 3–5 toxicity, discontinuation, QoL and treatment exposure require primary report/supplement extraction before full clinical signoff. Missing rates are not imputed.

Interpretation and limitations

Mature OS supports neoadjuvant chemoimmunotherapy in eligible resectable NSCLC. Extract full grade 3–5 toxicity, surgery completion, discontinuation and QoL from the full report.

Distinguish prespecified from exploratory subgroups, mature from immature endpoints, and superiority from non-inferiority. Verify current guidelines and individual eligibility.
